Optimaix-3D scaffolds are highly purified and can therefore be seeded in the dry state without previous washing with medium (preferred method). If required, the scaffolds can also be seeded in the wet state, but this will normally result in a reduced initial cell penetration.
Optimaix-3D scaffolds are characterized by a rapid and effective uptake of the cell suspension due to the capillary action of the 3D-structure. When applying the first drops of the cell suspension, a slight shrinkage of the scaffolds can be observed. This shrinkage will recover immediately when additional cell suspension is added.
Two procedures can be used for seeding the scaffolds, i.e. the ‘drop-on’ and the ‘drop-in’ methods (see below). After cell seeding, allow sufficient time (suggested minimum of 2 hours) for cell adhesion to take place before adding additional medium.
Drop-on cell seeding
This method allows control over the exact number of cells to be seeded on the scaffold. During the ‘Drop-on’ seeding method, a defined volume of the cell suspension is pipetted onto the scaffold to achieve near-complete rehydration.
Using the Drop-on-method, a slightly uneven distribution of cells within the scaffolds is obtained.
Drop-in cell seeding
This method will, in most cases, give an efficient, deep, uniform and homogeneous distribution of cells during seeding. In order to obtain a uniform cell distribution within the scaffolds, it is recommended to drop the scaffolds into an excess volume of freshly mixed cell suspension. After a few seconds, complete rehydration of the scaffold is achieved, which is associated with a highly efficient and uniform seeding.
Optimaix-3D scaffolds provide a unique 3-dimensional matrix for in vitro and in vivo investigation. As a result of the patented manufacturing process, the highly orientated and porous 3-dimensional scaffold offers a guidance structure that supports substantial cell migration, proliferation and maintained viability.
Optimaix-3D scaffolds have proven to be compatible with a wide range of cell types including stem cells (embryonic stem cells, bone marrow derived stem cells and adipose tissue derived stem cells), differentiated primary cells (e.g. fibroblasts, chondrocytes, adipocytes, smooth muscle cells, olfactory ensheathing cells, astrocytes and Schwann cells) as well as with a number of cell lines. Optimaix-3D scaffolds are stable under normal cell culture conditions and have typically been used for culturing periods of up to three weeks. In in vivo investigations, cell-seeded Optimaix-3D scaffolds have been demonstrated to be biodegradable in the absence of any significant inflammatory reactions. Furthermore, Optimaix-3D scaffolds did not induce immunological reactions.
